چکیده

The effects of free fatty acids on rat liver phosphoenolpyruvate carboxykiuase (EC 4.1.1.32), pyruvate carboxylase (EC 6.4.1. I), and glucose 6-phosphatase (EC 3.1.3.9) were determined to evaluate the suggestion that free fatty acid inhibition of key glycolytic enzymes constitutes a specific inhibitory effect. Activities of ah of these gluconeogenie enzymes were found to be markedly inhibited by long chain free fatty acids as well as by palmitoyl coenzyme A. The manner of the inhibition of gluconeogenic enzymes was indistinguishable from that reported for the glycolytic enzymes. It is therefore concluded that such effects in vitro of free fatty acids or long chain acyl-CoA esters on glycolytic and gluconeogenic enzymes may not be relevent for the regulation of metabolism in Go.

Inhibition of enzyme activities by free fatty acids.

The inhibition of several enzyme activities by free fatty acids has been examined. The inhibition of rat liver microsomal linolenate-activating enzyme activity by linolenate was found to be due to the inactivation of enzyme by substrate fatty acid. Stearoyl coenzyme A was also found to inhibit linolenate-activating enzyme. Likewise, microsomal glucose 6-phosphatase was inhibited by oleate; the ...

Regulation of the activity of acetyl coenzyme A carboxylase by palmitoyl coenzyme A and citrate.

Pahnitoyl-CoA (100 PM), in the presence of albumin (24 mg per ml), inhibited the incorporation of [14C]citrate into fatty acids in a cytosol fraction of chick liver and inhibited the activity of acetyl-CoA carboxylase purified from chick liver. Under similar incubation conditions, pahnitoyl-CoA (ZOO PM) did not inhibit fatty acid synthetase, ATP-citrate lyase, malic enzyme, NADP-linked isocitra...

Regulation of fatty acid oxidation in skeletal muscle.

Researchers using animals are beginning to elucidate the control of fatty acid metabolism in muscle at the molecular and enzymatic level. This review examines the physiological data that has been collected from human subjects in the context of the proposed control mechanisms. A number of factors, including the availability of free fatty acids and the abundance of fatty acid transporters, may in...

Reversible inhibition of mitochondrial adenosine diphosphate phosphorylation by long chain acyl coenzyme A esters.

Palmitoyl coenzyme A instantly and reversibly inhibited the electron transport-linked phosphorylation of ADP coupled to the oxidation of various substrates by rat heart mitochondria. The inhibitory effectiveness of pahnitoyl-CoA was dependent on mitochondrial as well as ADP concentration. Removal of palmitoyl-CoA, such as by its oxidation following (-)-carnitine addition, led to a restoration o...
